Overview

The Absolv module is a product of cooperation between Pharma Algorithms and Prof. M. H. Abraham.
Absolv Equations panel enables the user to calculate various solvent-solvent or gas-solvent partition coefficients from more than 100 predefined Abraham type equations may be calculated. Alternatively, any custom equations using predicted Abraham solvation parameters of a solute may be entered.

Module interface

Absolv Equations Screenshot.png
  1. Values calculated using selected equations
  2. Toggle the result of a corresponding equation invisible in the Results Pane
  3. All solvation equations are grouped according to their type. Switch between these groups by selecting an appropriate tab
  4. Name of a solvation property
  5. Equation used to calculate a particular property
  6. Comments on a corresponding equation
  7. Literature references
  8. Scroll through all available equations in the selected group
  9. Toggle the result of a corresponding equation visible/invisible in the Results pane
  10. Click to open the Equation Manager window

Equation Manager

Equation Manager allows defining new and editing or deleting the existing custom Abraham type equations as well as viewing all the predefined ones.

Note: The predefined internal equations can be neither altered nor deleted.

Equation Manager.png
  1. Select the equation’s category
  2. Select an individual equation
  3. Edit a custom equation or view the preset one
  4. Create a new custom equation
  5. Delete the created equation
  6. Close the Equation Manager window
Edit Equation.png
  1. Enter the name of the equation
  2. Enter the value of the intercept
  3. Enter a coefficient of the corresponding solute property in the formula
  4. Choose between the available H-Bonding basicity parameters (B or Bo)
  5. Select the appropriate cavity effect and dispersion interactions descriptor (L or V)
  6. Select the category of the equation from the drop-down list or enter a name of the new one
  7. Enter comment
  8. Enter a literature reference

Newly entered equations appear under the corresponding category in the Equation Manager and Equations pane:

Technical information

Calculated quantitative parameters

Parameters calculated by Absolv module represent the following properties of a solute:

  • H-Bonding acidity - α2H (A)
  • H-Bonding basicity - β2H (B) and β2O (Bo)
  • Partitioning coefficient between gaseous phase and hexadecane - log L16 (L)
  • Polarity / polarizability - π2H (S)
  • Excessive molar refraction - R2 (E)
  • McGowan characteristic volume - Vx (V)

Reference database

Absolv\Absolv DB module contains a browsable database of human intestinal absorption & bioavailability. The database consists of 5004 compounds with experimental values of solvation parameters determined by Prof. M. H. Abraham and coworkers. The main literature references where the values comprising the Absolv DB were presented include:

  • Abraham, M. H., Whiting, G. S., Doherty, R. M., and Shuely, W. J., J. Chromatogr., 1991 (587), 213-228.

Absolv DB module provides powerful searching and filtering capabilities, enabling the user to search compounds by name, by structural similarity, as well as limit the output to molecules possessing parameter values in the specified range or described in a particular publication.
